Starting to Paint in PS 7, but really lost. HELP!

Hello, all. I have been trying to learn as much as possible and practice as much as possible, but here I am. After a week, I finally got my line art down. Here it is:



I want to digitally render this in PS 7, but I would rather paint it than digitally color it like a typical comic book. I'm lost on how to get there. Do I ink it first, or will the pencil lines suffice? I am using a mouse and not a tablet, so that seems harder than I had hoped. Can someone give me a quick outline of how to proceed? I got my books and refence material, but...

I expect that I will do this in layers.
Background first? Do I paint over the line art? Multiple layer? Normal layer? Any help is appreciated.
